1. Indications

VONOPIN 20 mg may be prescribed for:

  • Erosive esophagitis — healing acid-related damage to the esophagus
  • Relief of heartburn associated with erosive esophagitis
  • H. pylori infection, when used together with appropriate antibiotics
  • Other acid-related conditions according to the locally approved product information.

2. Benefits / Pharmacological Action

  • Strongly suppresses gastric acid secretion

3. Usage / Dosage

For healing erosive esophagitis:
20 mg once daily for 8 weeks is the recommended adult dose in the current U.S. labeling.

For H. pylori infection: vonoprazan 20 mg is used with antibiotics. Regimens include:

  • Vonoprazan 20 mg + amoxicillin 1,000 mg + clarithromycin 500 mg, twice daily for 14 days, or
  • Vonoprazan 20 mg twice daily + amoxicillin 1,000 mg three times daily for 14 days.

Important: The H. pylori regimen should be prescribed by a doctor; do not use vonoprazan alone to treat H. pylori.

4. Contraindications

Do not use vonoprazan in patients with:

  • Known hypersensitivity/allergy to vonoprazan
  • Previous serious allergic reaction to the medicine
  • Concomitant use of rilpivirine-containing products.

5. Side Effects

Possible adverse effects include:

  • Diarrhea
  • Abdominal pain/discomfort
  • Nausea
  • Abdominal bloating
  • Constipation
  • Headache
  • Gastritis
  • Changes in liver enzymes

Rare but serious reactions can include severe allergic reactions, kidney inflammation, severe skin reactions, and C. difficile-associated diarrhea.

6. Warnings & Precautions

Use with medical supervision in patients with:

  • Liver disease
  • Kidney impairment
  • Low magnesium or other electrolyte abnormalities
  • Long-term acid-suppression requirements

Persistent symptoms despite treatment should be evaluated because improvement with acid suppression does not completely exclude gastric malignancy.

Long-term acid suppression can also be associated with problems such as vitamin B12 deficiency, low magnesium and gastric fundic-gland polyps.

7. How to Take

  • Take orally as prescribed.
  • It can generally be taken with or without food.
  • Take at approximately the same time each day.
  • Swallow the tablet as directed; do not alter/crush it unless the product’s specific instructions permit this.
