ONOPIN contains vonoprazan, a potassium-competitive acid blocker (P-CAB) that strongly and continuously suppresses gastric acid secretion.

1. Indications

Vonoprazan may be used for:

• Gastroesophageal reflux disease (GERD)
• Maintenance of healed erosive esophagitis
• Relief of heartburn associated with GERD
• Acid-related gastric/duodenal conditions, depending on the approved indication in the country
• Prevention of recurrence of certain ulcers associated with NSAID or low-dose aspirin use in some approved indications
• In combination with antibiotics for H. pylori eradication.

• Supports healing and maintenance of erosive esophagitis

3. Usual Dosage

For adults, the 10 mg strength is commonly used for:

• Maintenance of healed erosive esophagitis: 10 mg once daily, up to 6 months.
• Non-erosive GERD heartburn: 10 mg once daily for 4 weeks.

For active erosive esophagitis, the FDA-approved dose is generally 20 mg once daily for 8 weeks, rather than 10 mg.

H. pylori treatment requires combination therapy with antibiotics and uses vonoprazan 20 mg—not 10 mg—under the current U.S. regimen.

4. Contraindications

Do not use vonoprazan in patients with:

• Known allergy/hypersensitivity to vonoprazan or its ingredients
• Concomitant use of rilpivirine-containing products under the FDA labeling.

5. Side Effects

Common side effects may include:

• Diarrhea
• Abdominal pain or discomfort
• Nausea
• Abdominal bloating/distension
• Gastritis
• Headache
• Dyspepsia

The exact adverse-effect profile can vary according to the indication and treatment combination.

Seek medical attention for severe allergic reactions, severe/persistent diarrhea, significant skin reactions, or symptoms suggesting kidney problems.

6. Warnings & Precautions

Long-term or inappropriate acid suppression may be associated with:

• Vitamin B12 deficiency
• Low magnesium levels
• Increased risk of C. difficile-associated diarrhea
• Bone fractures, particularly with prolonged use and other risk factors
• Acute tubulointerstitial nephritis
• Fundic gland polyps with long-term treatment
• Severe skin reactions in rare cases.

Patients with liver or kidney impairment may require dose adjustment or additional precautions.

7. Usage / Administration

• Take once daily when prescribed.
• Can generally be taken with or without food.
• Swallow the tablet whole.
• Do not chew or crush unless the specific product information permits it.
